No fluorescence was observed in the region 227 between the pupal cuticle and the apical surface of the epithelial cells in Ore-R pupae (Fig. 6EF) . We also observed puncta in very young pupae (20 237 hr awp) prior to the detachment of the epithelial cells from the pupal cuticle (Fig. S6B) . These animals showed a large 253 number of fluorescent puncta present in the space between the pupal cuticle and the apical surface of 254 the epithelial cells ( Fig 6GHI) .
