French and Roy (1990) constructed a dual-recombinant baculovirus containing genes that encode VP3 and VP7 and their expression in insect cell culture resulted in the synthesis of large quantities of VP3 and VP7 and the concomitant formation of core-like particles (CLP) that lack RNA. Ovine polyclonal antiserum was raised to core particles of BTV-1 isolated from virus-infected cells and purified by equilibrium density centrifugation in cesium chloride (personal communication, B.T. Eaton). The number of crude CLP per negative film was determined by the ISEM method and the concentration of particles calculated by multiplying the number of CLP by the capture efficiency.
