To test the hypothesis that a CfN DNA-protein complex was responsible for the transcriptional block, we replaced the wild-type CENG sequence with a CEN6 sequence containing a single-nucleotide point mutation (CDEIII-15C) in the central element of the palindrome of CDEIII (CCG). Of 34 cff mutants screened (see Experimental Procedures for list), 7 were identified as putative kinetochore mutants because they produced an intermediate level of blue colony color between the levels of the CTF+ strains carrying the wild-type and mutant CEN reporters. ./cache/cord-270082-byxd4o4m.txt ./txt/cord-270082-byxd4o4m.txt
