Strips were incubated with hybridoma supernatants diluted 1/10 in primary buffer (TBS/Tw, 1 mM EDTA, with 1 mg HSA and 100 μg normal human IgG (hIgG) added per ml), or with the parental rat serum 1/500. The wells were then blocked with HSA, 1 mg/ml TBS, washed thrice with TBS/Tw, and incubated with serum samples diluted 20-fold in MAp19 buffer (TBS/Tw, 1 M total NaCl, 10 mM EDTA, containing 100 μg ΔhIgG (heat-aggregated hIgG); Beriglobin, from ZLB Behring GmbH, incubated 30 min at 63°C and centrifuged 10 min at 3,000 g to remove large aggregates) per ml, and 100 μg normal rat IgG (Lampire) per ml) o.n. at RT.
